Does anyone know if it's possible to have more than 256 columns in Excel?

AFAIK it can't be altered from the standard 256 columns.
I haven't yet seen a spreadsheet though with this number of used columns.
Can you rotate your data (limits you to 256 rows though)?
Can you split your data into multiple sheets?

If you have 256 columns, that's a sign of terribly poor design, rather than anything else. Such a spreadsheet can't really be used by a human. What are you trying to do?
