I have a pair of Shimano MP66 shoes with SPD cleats. Can they be used with road bike SPD pedals?

Depends on which pedals you have. Although SPD pedals like the Shimano M520's are also used on road bikes, strictly speaking they are classified as MTB pedals. If on the other hand by "road bike SPD pedals" you are referring to Shimano SPD-SL pedals then the answer is no. You'd need SPD-SL cleats and adapters for the shoes (if available) to fit the 3 bolt SPD-SL cleats to the 2 bolt shoes that you have.
