    depends on the level of maths you are comfortable with,

    simple line, you can measure the y2-y1/(x2-x1) to get slope.

    another way would be to differentiate the line with resepct to the x axis and apply value for the determinant at the point of interest.

    But as mentioned above, your question is very vague....
